O.P. backing up software...
if i go to my computer > F: drive (after formatting my C: drive became an F: drive but it is the same !!!) > programs and files > (then find publisher )
and copy all the files from that directory to a file say on the desktop and burn it to CD

will i be able to reinstall publisher onto my PC again?

{im formatting a mates PC and dont have a copy of publisher i can install and i think she has it on there already}

Most likely not, as the program also creates regristry keys etc (in the most cases).

No!

Programs need to be installed.

When you install a program there are far more things that are copied to your hard disk than what you'll find in the programs directory. Many programs (almost all) install libraries too, alter registry settings, etc. etc. Without all this your program will simply not work (and/or may cause more errors in other parts of windows when run).

The only thing you can safely copy from one directory to the other (on another PC) are your own made documents. To backup the program itself, you need to backup your entire harddisk (simply speaking) and hope that the new computer has exactly the same hardware and set up as your original computer; otherwise things will not work either...

The best, and only safe, solution is to reinstall the program from scratch on the new computer.

is it also possible to keep the files after formatting, but then run the installer and repair it?

no because it wont detect an install of it , so it will prompt you to install it :P
