This method will only work if the sound card support full duplex mode. Note that not all sound cards support this.

Full Duplexing means the possebility to record and play at the same time.

Half duplexing is also possible. This means you can start recording and while doing that play something. But you can't start playing something and then start recording. (Half-duplexing is very rare though, and often makes no sense).

That would be a messy method and would bring down the quality of the sounds (because the output of the sound depends on your local volume settings and thus often over amplified and it also depends on your quality of devices and the Windows settings you use to play sounds).

If you want custom Plus! sounds from existing files, then use the "create sound from file" option, this will ensure you have the highest quality because nothing depends on your own Windows settings and hardware settings. The original file is used as a source (aka, you don't even have to have a sound device for this because the file is simply re-encoded, nothing gets played at all).

If you want custom Plus! sounds from external input, then use the "record sound from another source" and select your line-in, as this give you the best quality type of input (heck, it is especially made for this purpose). Doing this over a microphone again over-amplifies the sound on many motherboards.
